I've personally climbed this a few times on each route so know it quite well.
The most common way of climbing Lugnaquilla is by staring at Glenmalure. It's near Glendalough.
The best way to get to Glenmalure from Dublin or Bray would be to get onto the N11 heading south. Get off it at Kilmacanogue and head for the Roundwood / Glendalough / Laragh direction. Go through Roundwood, then Annamoe and to Laragh. At Laragh take the turnoff for Glenmalure / Rathdrum and fork to the right heading for Glenmalure. Drive over the hill and down into Glenmalure. At Glenmalure take a right at the junction (beside the Glenmalure Lodge Pub) and continue down the valley towards then end (its a cul de sac). There are several ways of climbing it from there.
The other way (easiest) of climbing Luqnaquilla is from the Glen of Imaal beside (outside) the Army Artillery Range. You must check with them in advance to see if they're firing that day. You can phone them at: 045-404653 or 045-404626. To get there take the N81 from Dublin through Tallaght, Blessington, and south towards Baltinglass. Take the exit for Donard and then into the Glen of Imaal to the Army Information and Advice Centre where you should start.
Whatever way you're doing it I would strongly reccommend buying the Ordnance Survey 1:50000 Sheet 56 Map.
Good luck!
